Antibiome Center, Department of Pharmaceutical Chemistry UCSF

The Antibiome Center is seeking a highly motivated candidate to join our quick paced and multi-talented team to develop antibodies at the proteome-wide scale. The successful candidate will be responsible for molecular biology, generation of antibody-display library, protein-antibody expression and purification, and protein-antibody validation. The candidate will have opportunities to learn state-of-the-art techniques and automation systems. Our laboratory is highly collaborative and is engaged in numerous academic and industrial collaborative projects.

The Antibiome Center is part of the Recombinant Antibody Network (RAN), a collaborative consortium consisting of laboratories at the University of California San Francisco (Dr. Jim Wells), University of Chicago (Dr. Tony Kossiakoff), and the University of Toronto (Dr. Sachdev Sidhu). We are committed to generate recombinant antibodies at the surface proteome-wide scale using high throughput phage display approaches. Our long history of protein engineering expertise also enables the development of bio-therapeutic antibodies in the immuno-oncology field.

Primary responsbilities:



  • Cloning recombinant protein and antibody constructs (including reformatting to scFv, bispecifics) using a variety of molecular biology techniques including Polymerase Chain Reaction, Gibson assembly, DNA purification, and transformation
    -Antibody display library generation (phage, yeast, or mammalian) including library design (Kunkel mutagenesis, Golden Gate Assembly, Lentivirus production)
  • Protein expression from E. coli and mammalian sources (HEK293 and CHO cells)
  • Purification of proteins using various chromatography techniques such as affinity with Protein A, IMAC with Ni-NTA, and IEX
  • Characterization of proteins by variety of techniques including SDS-PAGE, ELISA, SEC, and Western Blotting


Required Qualifications:



  • Junior Specialist- A B.S. degree (or equivalent degree) in biochemistry, cell biology, or relevant life sciences field or at least four years of research experience
  • Assistant Specialist- A master's degree in biochemistry, cell biology, or relevant life sciences field or a bachelor's degree with 3 or more years of research experience.
  • Associate rank- a master's degree (or equivalent degree) in biochemistry, cell biology, or relevant life sciences field or five to ten years research experience.
  • Candidates must meet the required qualifications at the time of appointment. Candidate's CV must state qualifications (or if pending) upon submission.


Preferred Qualifications:



  • Expertise in display library generation and selection.
  • Experience with Next-Generation-Sequencing techniques and analysis.
  • Familiarity with mammalian tissue culture techniques including transfections, cell line maintenance, recombinant protein expression.
  • Proficiency with advance biophysical such as Biolayer Interferometry (BLI), Surface Plasmon Resonance (SPR), poly-reactivity assays, HPLC chromatography
  • Data analysis experience using programing languages such as R or Python.
  • Junior/Assistant Specialist- at least one year of wet laboratory experience
